You can use the test points or solder a connector on. They are connected together.
The test points could be handy if you design your own carrier board and you want to use the UART in your design. You can use pogo pins to make a connection without having to solder to the CM5.
The connector option is probably better if you want to debug during development. You don't really want to have to solder on to every CM5 on a final product.
